Analyse der Proteinkristallisation von Lysozym mithilfe von Statischer Lichtstreuung

Abstract

This paper is about the technique of static light scattering (SLS). In general, a probe inside a so-called scattering chamber is enlightened with a monochromatic laser beam. Around the scattering centre are 18 Detectors. The intensity collected at each of the detectors lets us make statements about the size and structure of the probe inside the chamber. The final destiny of this work is to give a brief insight of the process of protein crystallisation, which is still a very unknown procedure in the field of scattering. Proofing certain concentrations of the two components of a protein crystallisation (NaCl and Lysozyme; [4] p. 35f.) and giving a broad statement about the phases of the growth of protein crystals would be a good target for this work. To understand and learn the procedures of proper sample preparation and measurement, we use silica nanospheres varying in diameter from 100nm-500nm. Using this knowledge we can extract certain time points of the crystallisation and predicate about the stage of growth of the time period (1-2min). The general hope is to see phases like the first nucleation, the period of growth and the first finished crystals.
