Detektion der α4-, α5- und δ-Untereinheit von GABA-Rezeptoren im Thalamus des Mäusehirns mittels Immunfluoreszenz

Abstract

The goal of the research was to detect the GABAA receptor in the thalamus of the mouse brain. This was carried out by using the indirect Immunofluorescence method to indicate its α4, α5 and δ subunits. For this purpose, different series of experiments with tissue samples from the mouse brain were performed. Besides the tissue samples from wild type mice, knockout mice with deactivated target proteins were used to prove that no other than the demanded structures have been detected.
During the experimental phase of the project various parameters have been optimized to improve the quality of the resulting images.
The fluorescence images showed light spots proving the presence of α4, α5 and δ subunits of the GABAA receptor in the mouse brain. Nevertheless the target structures were also detected in knockout mice, which indicates faults in the process of deactivating the subunit.
The aim of the project was to prove the presence of GABAA receptors in tissue samples by means of the Immunofluorescence method. This objective has been met: A working protocol for the detection of α4, α5 and δ subunits in the thalamus of the mouse brain has been found.
